介绍一种感受态细胞，一分钟转化质粒。

上周买了一种感受态细胞，昨天试用了一次。我很满意。对于小质粒（<6kb），整个转化过程只用1分钟。把细胞在流水下熔化后，加质粒，混均，然后直接铺到室温或者37°C的平板上即可（或者在42°C 30秒，铺到4°C的平板上）。对于大质粒（>6kb），在加入质粒后多加一步冰上孵育1－5分钟即可。转化效率很高。我用的是最便宜的一种，据说pUC19的转化效率是每微克质粒一千万转化子。价钱比一般公司卖的感受态细胞还便宜一些。对于象我这样不想花两小时转化细菌的人，值得一试。生产这种细菌的公司是一个台湾省的公司，大家也来支持国货一把。定货信息如下：

I ordered some ECOS Competent Cells produced by a Taiwan province, PRC based company called Yeastern Biotech. I am glad that I tried this product because it is efficient and time-saving. The transformation protocol omits the time consuming incubation steps. For small plasmids (<6kb), the protocol takes 1 minute. The cells are thawed under running tap water for about 20 seconds, after adding the plasmid, the cells can be plated directly onto a warm (R.T. to 37°C) plate or be heat-shocked at 45°C for 15 seconds and plated onto a pre-chilled plate. For large plasmids, add a 1-5 min. ice incubation step will increase the efficiency. The company says the transformation efficiency of their cheapest cells are about 10 to 100 million transformants per µg of pUC19. After trying the cells yesterday, I have to say that the efficiency is really high. And I like it that it can save me almost two hours for transformation. 今天离人生完整更进一步。晚上下班回家，7点左右。从公汽走下来，很少有的只有我一人。走在回家的路上又经过了那个小公园，公园里的灯给人一种超现实主义的感觉。我兴致勃勃地走着，突然发现背后有两个黑影闪过来。快得很不正常。一人从后面把我抱住，对另一人说：“Get the Wallet”。另一人上来搜索一番。摸到右边裤包里的钥匙，扔了出来。如果我背了包，几乎从不把钱包放在衣兜里。两人什么都没有找到，很郁闷地消失了。居然也不顺便翻一翻我的背包 (今天刚取出的20块钱），抱我那人还转到前面来让我看清了脸。难道是黑人小伙子们看到了我沧桑的脸吓到了？往前走了几步，一辆汽车突然发动起来，不知道车上的人是这两人的同伙呢，还是正好坐在车里偶然观赏到了一次突发事件。反正我也没看清车牌。 人说久走夜路必撞鬼，必然要发生的事情终于发生了，只是我没有想到等了近一年。我从来不担心钱包被抢，各种卡都可以补办，只要不遇到穷凶极恶的上来不问青红皂白就给你来一刀或者请你吃一颗花生米的就问题不大。回想起来这两人还不坏，动作轻柔，没有搞到好处也没有报复性地给我几拳，让我的这次体验显得很不真实，就象我在回家的路中途停留了一次，对之前之后没有丝毫影响。我经常同老婆开玩笑，说黑人的皮肤是天然的夜行服。他们身着深色的衣服，在夜色中来无踪去无影。如果不是我捡起地上的钥匙，恐怕我会以为自己在梦中。

I didn't have time to enjoy the city's better side. I'll check it out next time.

那天我偷偷给老婆说，自从有了猪儿八后，我看她就觉得肢体巨大，手脚身脸和宝宝比较起来，完全没有那个玲珑劲，让我特别不爽。皮肤更不用说了。亮堂堂的小屁股，摸上去简直就是享受：又细又软又有肉头。老婆给我说，她也有同感。人长那么大，又没有美感，又没有用处。呵呵，这个猪头。

Traffic Jam can be beautiful at night. The blue and green neon lights, the long lines of red break lights at the back of the cars and the yellow head lights at the front, and the skyline of the city forms an unique combination. It's even better when I am on the metro trail. Well, of course, I didn't mean that I enjoy watching people stuck in the traffic. Different prospective gives different view. The same as life. When we look at others' lives and we think they're wonderful, how likely those who live in them are actually suffering?

太乙真人给哪吒重塑肉身的时候用了莲藕，所以哪吒后来生得粉嫩粉嫩的。看着我们亮堂堂肥肥的大腿，我终于明白了。我们猪肉小朋友腿上一褶褶的肉，不正象莲藕一样么？

大家在用PubMed搜索文献的时候，不知道有没有想过National Library of Medicine（NLM）做为一个图书馆的真实存在。NLM就坐落在NIH内，面向大众开放。除了收藏医学相关图书之外，还有博物馆的功能，有各种同医学相关的展览，供公众参观。没有机会到NLM一游也没有关系，因为很多展览在网上也可以看到。最近媒体中流行的展览是“Magic and Medicine in Harry Potter”，很有趣的小展览(网址： http://www.nlm.nih.gov/exhibition/mandrakes/），不知道Prof. Snape看后作何感想。

我们宝宝现在可以清楚的发出三个音了：哈，吧，啊。我于是对他讲：“你有什么事情要告诉老爸老妈，不能老是用哭的方式讲，可以用这三个音嘛。哈就是要吃了哈；吧就是要拉巴巴拉尿尿了；啊就是胃不舒服要拍拍啊。”说了半天，他一点反应也没有，还是在那里哈哈哈的叫。我也是太拔苗助长了，他要是真能说话了，我该吓一大跳。 Baby JJ师兄说要当我们猪儿八的粉丝。我决定专门为他给猪猪猪起一个绰号：“猪肉”。现在我正式宣布，Baby JJ师兄是“猪肉粉丝”了。 今天发一张亮堂堂昨天的照片： 一只耳朵， 左耳， 亮堂堂的左耳。 这只耳朵长在头的左边， 上面长着浓密的胎毛。 耳朵不会动， 因为它是猪耳朵， 猪耳朵不会动， 会动的是狼耳朵。

我发现猪猪猪对音乐的敏感性不是一般的好。今天哭闹的给他放Ignaz Moscheles的Characteristic Studies, Op.95, 他立即停止哭闹，听得很认真的样子。而且眼睛可以跟随猪爸打着拍子的手指运动。头也转去，耳朵向着音箱的方向。猪爸给猪妈说，宝宝如果长大后成为音乐家，你喜不喜欢？猪妈说好啊。亮堂堂立即就笑起来。

人奶里的维生素D比较容易被破坏。所以如果是母乳喂养，而宝宝没有机会晒到太阳，就有缺维生素D的可能。一般有三种方法补充：1.妈妈补VD，2.晒太阳，每天5-10分钟足够；3直接给宝宝补充。我们家猪妈一直在用prenatal的维生素（prenatal的维生素可以吃到哺乳期，就要是补充叶酸，钙，铁，VD等维生素）。因为冬天来了，娃娃出门的机会少，所以上周儿科建议补充维生素ACD和铁，如果宝宝长得快，还可以补一些族维生素。不过我们查了查资料，如果妈妈补了铁，母乳里的铁应该足够了。而且补铁剂，宝宝吃了会不太舒服。所以我们决定不补铁了。Emfamil的Tri-Vi-Sol有维生素ACD，（Tri-Vi-Sol with Iron是了铁的），还有一种是Poly-Vi-Sol，有维生素ACDE及B族维生素。我们买了Poly-Vi-Sol. 看网上的父母们说，这个东西不太好吃，很多小孩吃不下去，要吐要闹。我们今天早上（早上宝宝一般不闹）在他有一次吃奶前喂了他吃。果然吐，不过还是吃下去不少。然后猪妈就给猪儿八喂奶，冲下去了。如果是吃配方奶，应该就不用补充维生素了。

宝宝对音乐比较有感觉。能分辩出好坏了。我给他听不同的版本的音乐，他仿佛都清楚。中文歌曲我也给他听，其中他比较喜欢的是吴雁泽唱的《草原上升起不落的太阳》。我现在正在试验，每天给他放不同风格的音乐。不过到现在还主要是中西方古典音乐和一些歌曲。比较吵的现代派音乐及摇滚乐还没有试过。等他大一些吧。不过他对吵一些的音乐到也不排斥，上回我们看的一部电影里的音乐也吵的，节奏感比较强，猪爸抱着他跳舞，亮堂堂也是很高兴的样子。

好久没有做饭了。今天是感恩节，所以做了一顿西餐吃：1. Picnic Pork Shoulder Roast（烤的猪肘子，因为实在是不想吃火鸡）；2. Tilapia Fillet Roast seasoned with a blend of basil, allspice, ginger powder, paprika, and olive oil; 3. Butter garlic broccoli stir; 4. Pumpkin rice with Italian seasoning; 5. Quince apple pomegranate sweet with cinnamon; 6. Molten Chocolate Cake.

昨天晚上做实验回家比较晚，回到家的时候已经差不多9点了。猪妈说猪儿八从4点闹到9点。一听到我开门的声音，就直往外瞅。看到我回来，立即就不闹了，乖乖地吃奶睡了。好象我在家的时候，亮堂堂总是比较乖。这让猪妈比较郁闷。说猪爸只看到猪猪猪可爱的一面，没有看到耍混的一面。这个小小两面派！记得有一天给猪妈打电话，猪妈说在给黄八斤喂奶，但是他闹着不吃。我说："让儿子接电话“。儿子接后我说：”息息，乖乖吃奶，不然爸爸回来打屁股！“他听到后果然止哭乖乖吃奶。我不知道小孩子到底有多明白整理，不过娃娃心理面清醒得很，只是说不出来罢了。

今天是感恩节，放假在家。一般来讲，明天也没人去上班（购物时间），所以我也不去了。这一周连放四天。现在我是理解到了，在家的时候比上班累。就要是带娃娃不象做实验。做实验失败了可以重来，今天做不完可以以后再做。带娃娃可没有这样的机会。象我这样睡觉如猪的人，自从宝宝回了家，就没有睡过一个整觉。做父母的辛苦，只有自己设身处地后才能真正了解。

三十三天大的宝宝俨然是大娃娃模样。人人都说我们家宝宝乖，是真乖，还是我自己的偏见？

达赖喇嘛还没死，就急着想去投胎了。这个可悲的小丑。不知道转世的灵童会不会说英语。把他放到白宫他还能记得同小布什谈过话么？

关于木瓜丰胸说

上周在MITBBS上看了一人用木瓜丰胸未遂的惨痛帖子。唉。男人女人都有想变大的地方，可惜这些地方的大小都由基因决定。事实就是事实，怨天尤人也没有用。做手术，那是假的借的；各种偏方秘方，根本没有有用的。

木瓜下奶的效果到是不错。能下奶自然可以丰胸，不过那丰的是哺乳期妇女的胸。过时不候。别的人，对不起，没有天时地利人和：既没有黄体分泌激素，又没有嗷嗷待哺的婴儿刺激。这个胸，怕是丰不起来。还是看着人家的真真假假的东西流口水比较实在。

猪猪猪昨天很勇敢。打疫苗很痛，他只哭了两声。不到两周，猪儿又长了1镑9盎司，平均一天1点8盎司，就跟催肥的一样。这段时间他火比较重，脸上长了不少红籽籽。我于是叫他黄麻子。昨天去看的那位儿科医生特别会弄娃娃。亮堂堂在他手中非常听话。我们在边上也学到几手：轻轻摸摸脸，点一点鼻子和小巴，揉一揉脖子。宝宝居然一点也不哭，不象前两次。

猪妈开始教黄麻子坏话了。这还要怪武机长。猪妈抱着娃娃睡觉，嘴里哼的是摩尔斯电码：滴滴哒滴 滴滴哒 哒滴哒滴 哒滴哒。唉，从小就不学好，长大了怎么得了。
